They will help but I would only do a few and make them quality as opposed to "doing a ton".
If you really want to bust out a good 400 time start training with hills. Do repeats of varying length and effort levels.
You also mention doing a 10 miler which in my opinion is not necessary for a quarter miler. You'll have good endurance but you could be far more efficient keeping your long run under 8 miles, doing 2-3 moderate to easy runs, and then hills and speed work.

Only If you add in other strides as well. Running a bunch of 100's might make your fast-twich muscles stronger, but they won't give you the endurance you want for the 400. I'd add in 1 and 2 mile runs and do the 100's afterwords that was you can work your fast twich muchles when you are tried and improve your overall endurance.

I think you are mixing your workouts. The 10 mile run should be aerobic base building. Your 100's are for speed, the two workouts generally don't get mixed. I would suggest a regular speed session and leave the long run a long run.

i think that it would because it helps you build up your speed but you should also try to sprint 40's and 50's to help you
